Cannabinoid receptor 1
The CNR1 gene at 6q15 codes for the 472 amino acid protein cannabinoid receptor 1. Three isoforms are potentially produced by alternative splicing although one is so short at 38 amino acids its role is unclear. Excessive stimulation of cannabinoid receptor 1 is associated with obesity and impaired metabolic function.It is localised in the periphery in brown adipose tissue as well as being expressed in the CNS. It is known that macrophage phagocytosis is mediated by CB1 not CB2. Some genetic variants of CNR1 are associated with obesity-related phenotypes in men. Rimonabant a drug developed to block the receptor was withdrawn due to the fairly predictable systemic side effect of depression.
