Casein kinase

Casein kinase is actually a group of human enzymes that phosphorylate a large number of proteins as part of the Wnt pathway. They include:

  • Casein kinase I isoform alpha ( CK1, CKI-alpha, EC 2.7.11.1) coded for by the gene CSNK1A1 coded for at 13q13 which phosphorylates CTNNB1 on 'Ser-45' and interacts with axin complexes. It binds ATP and nucleotides
  • Casein kinase I isoform delta (CKId, CKI-delta, EC 2.7.11.1) coded for by the gene CSNK1D coded for at 17q25 which is a central component of the circadian clock
  • Casein kinase I isoform epsilon (CKIe, CKI-epsilon, EC 2.7.11.1) coded for by the gene CSNK1E at 22q12-q13 another component of the circadian clock and phosphorylates DVL1 in the Wnt pathway.
  • Casein kinase I isoform gamma 1 (CKI-gamma-1) coded for by the gene CSNK1G1 at 15q22.1-q22.31
  • Casein kinase I isoform gamma 2 (CKI-gamma-2) coded for by the gene CSNK1G2 at 19p13.3
  • Casein kinase I isoform gamma 3 (CKI-gamma-3) coded for by the gene CSNK1G3 at 5q23
  • Casein kinase II which has subunits:
    1. Casein kinase II subunit alpha 1 (CKII, EC 2.7.11.1) coded for by gene CSNK2A1 at 20p13 which is known after UV radiation to phosphorylate'Ser-392' of the p53 tumour suppressor gene.
    2. Casein kinase II subunit alpha 2 coded for by gene CSNK2A2 at 16p13.3-p13.2.
    3. Casein kinase II subunit beta (CK II beta, phosvitin, G5a) coded for by gene CSNK2B (G5A) at 6p21.3.
