Inclisiran

Inclisiran is a modified siRNA which has been conjugated to a triantennary N-actylgalactoseamine (GalNAc)[1]. This allows uptake after binding to the hepatocyte asialoglycoprotein receptor and after intracellular transport hybridises to PCSK9 gene generated mRNA (the guide strand) and with its passenger strand designed to interact with the RNA-induced silencing complex (RISC) stops mRNA transcription. A single subcutaneous injection reduces LDL-cholesterol for over 6 months[2].
