Serine/threonine-protein kinase mTOR is the key protein kinase in mTOR signalling coded by the MTOR gene at 1p36.2. The gene codes for a 2549 amino acid peptide which even autophosphorylates itself. A component of the mammalian target of rapamycin complexs (mTORC1 and mTORC2). With mTORC1 its binding to sirolimus inhibits the complex. With mTORC2 sirolimus needs very high concentrations to inhibit due to other components of the complex altering the binding site characteristics. It is expressed in highest levels in the testis, but is found in most tissues in the endoplasmic reticulum membrane, cytoplasmic side of the peripheral membrane protein, Golgi apparatus membrane, mitochondrion outer membrane and lysosome membranes.
