Programmed cell death 1 ligand 2 is coded by the PDCD1LG2 gene at 9p24.2 to produce a 273 amino acid peptide which is further processed into a key ligand for the programmed cell death protein 1 receptor that inhibits T cells. It is also involved independently in the costimulatory signal, essential for T-cell proliferation and production of interferon gamma. There are three isoforms, produced by alternative splicing being:
  1. PD-L2I - A single-pass type I membrane protein found on cell membranes particularly of heart, placenta, pancreas, lung and liver and more weakly expressed in spleen, lymph nodes and thymus
  2. PD-L2II - A single-pass type I membrane protein found on endomembrane system
  3. PD-L2III - secreted
