A guanosine analog hydrolyzed on absorption to aciclovir that herpes viral thymidine kinase converts to aciclovir monophosphate. This is then further phosphorolated by cellular guanylate kinase, and finally phosphoglycerate kinase, phosphoenolpyruvate carboxykinase, and pyruvate kinase to aciclovir triphosphate which competitively inhibits viral DNA polymerase by competing with deoxyguanosine triphosphate, for incorporation into the viral DNA. If incorporated into DNA, aciclovir triphosphate inhibits DNA synthesis by acting as a chain terminator.
